Is it considered unconventional or potentially problematic to contemplate elevating the floor of my garage or shop by utilizing pressure-treated (PT) 2x4s and oriented strand board (OSB)?

Sounds like too weak for any practical purpose.  OSB is poor as structural material.  

Of course, I wouldn't even consider it for canoe storage.

Only thing I'd use would be some old tongue and groove 2x6 for the flooring. The 2x4's if they are resting on the existing cement will work fine on 16" centers. No OSB for a garage floor. It's barely OK for residential flooring.

OSB for a floor in a garage is a bad idea even if it's laid directly on concrete. 100% it will absorb moisture and oils and warp and sag at best and collapse at worst. It's a cheap fiberboard. 

What is the purpose of this exercise and how do you use your garage?

Why not just re-surface with more concrete? There is a procedure for re-grading concrete that works fine if you are adding a couple inches(though some of the tools required are somewhat specialized)
