My brother gave me an old Sekine 10 speed (dunno what model) about 25 years ago that's done nothing but take up space since. At 330 lb I'm obviously far too heavy for the thing. However, with two small girls SWMBO is thinking that family biking on the KATY trail is coming soon. Would it be better to buy some sort of Mountain bike for myself OR put some appropriate tires and wheels on the Sekine for my lardass?

If you really want to spend some time riding find a bike that is comfortable. I don't know anything about Sekine, but if the ergonomics of it don't work for you, you wont be on it for very long.

You should be able to sell the Sekine reasonably easy - they are moderately popular right now, as are all old ten-speeds. It's a decent bike most likely, but you are right about the weight limit. It will be more of a PITA to put heavy-duty wheels and tires on the Sekine than buying a mountain bike built to sustain your frame.
